2’3’二硫代磷酸（酯）是3’3’-環(huán)磷酸腺苷(c-di-AMP)的異構(gòu)體類(lèi)似物。c-di-AMP是機(jī)體經(jīng)bacteria刺激后產(chǎn)生的第二信使，在哺乳動(dòng)物體內(nèi)具有很強(qiáng)的免疫刺激活性[1]。

該環(huán)狀核苷酸（CND）可誘導(dǎo)產(chǎn)生I型干擾素（IFN），進(jìn)而被內(nèi)質(zhì)網(wǎng)駐留型受體STING（干擾素基因刺激物）、TBK1（TANK結(jié)合激酶1）和IRF3（干擾素調(diào)節(jié)因子3）識(shí)別[2]。

人內(nèi)源性的CDN是由CGAS（環(huán)GMP-AMP（cGAMP）合酶）所產(chǎn)生的[3] ，由于其存在2’-5’, 3’-5’混合鏈?zhǔn)浇Y(jié)構(gòu)，因而2’3’-c-di-AM(PS)2 (Rp,Rp) 與STING的親和性要高于c-di-AMP與STING的親和性。2’3’-c-di-AM(PS)2 (Rp,Rp) 可激活所有人的STING等位基因以及小鼠的STING基因。

此外，該類(lèi)似物包含兩個(gè)硫代磷酸酯二酯鍵，可防止其被宿主細(xì)胞或循環(huán)系統(tǒng)內(nèi)存在的磷酸二酯酶降解[4]。

在體外實(shí)驗(yàn)研究中發(fā)現(xiàn)，與RP / SP二硫非對(duì)映異構(gòu)體或c-di-AMP相比，Rp, Rp二硫代非對(duì)映異構(gòu)體可誘導(dǎo)產(chǎn)生更多的I型IFN，并且還可以通過(guò)增加小鼠體內(nèi)2'3'-cGAMP來(lái)增強(qiáng)其抗腫瘤活性[3]。最近體內(nèi)研究發(fā)現(xiàn)2’3’-c-di-AM(PS)2 (Rp,Rp)能夠強(qiáng)烈激活腫瘤抗原特異性的CD8+T細(xì)胞并且能夠克服與PD-L1封閉聯(lián)合treatment中抗原引導(dǎo)的免疫耐受[5]。
